蛋白质的构象变化是其执行各种生物功能的基础,例如酶催化、信号转导和分子识别。这些构象变化通常由变构效应控制,即配体结合或突变等扰动在空间上远离活性位点,却能够影响蛋白质的功能。理解这种变构控制对于设计能够响应特定刺激而改变构象的蛋白质至关重要,例如荧光生物传感器。
传统的变构效应研究方法通常依赖于结构生物学技术,例如X射线晶体学和核磁共振波谱。然而,这些方法通常只能提供静态的结构信息,难以捕捉构象变化的动态过程。近年来,计算模拟方法,例如分子动力学模拟,已经成为研究蛋白质构象变化和变构效应的重要工具。然而,由于蛋白质系统的复杂性和计算资源的限制,全面探索变构控制机制仍然具有挑战性。
